According to Game Informer, the Big Sister was a Little Sister who grew up and came back to Rapture (which was in ruins, by the way)10 years after the first game with a bunch of kidnapped girls from the surface to make Little Sisters from. Basically, she's nuts about the Status Quo in Rapture, and if you upset it, usually by killing Big Daddies, she comes after you. Your warning is the Little Sister singing like you hear at the end of the trailer.
Wait a sec. So the big sister is a little sister that comes back to try and rebuild the little sisters?

If we go off this:

According to Game Informer, not just an earlier Big Daddy: the first Big Daddy, "made" before the ability to use plasmids was engineered out of them, no longer a slave to the little sisters, able to choose his own destiny. Awesome.
So somehow the first big daddy came back? And if the big sister loves big daddies so much, why would she try and attack you (filling the room with seawater wasn't exactly friendly)

I don't understand.

She doesn't love Big Daddies, she loves the Status Quo, what with the new Little Sisters going around getting Adam from corpses with the Big Daddies (old and presumably new). But, even if you're a Big Daddy, if you go around killing other Big Daddies to harvest/adopt (that's right, the good option is to adopt and defend Little Sisters while they go on their harvest trips, and you get a kickback of Adam for each corpse she harvests) Little Sisters, you upset that status quo, which pisses her off.

EDIT:Oh, and the first Big Daddy doesn't come back, he was always there, all 10 years. Despite Rapture falling into ruin, there are still people living there, including old big daddies.

Just to add to your post, the harvesting side mission is completely voluntary, which means if you decide to adopt a little sister, you can choose whether or not she harvest Adam at all or just carry her to a place where Tenebaum will most likely turn her back to normal.

I hadn't even heard about Tenenbaum turning them back. Was that in Game Informer? Maybe I just didn't read closely enough.

It wasn't, but I think that would be the logical conclusion since it's pretty much what you did in the first game, save the little sister and then she runs off to Tenenbaum.

Hmm, it strikes me, (without doing deeper research) that the statue in the underwater scene seems a bit religious in nature...

Apparently you'll be able to combined plasmids as well to form new ones, according to PC gamer at least.

I am really excited for this to come out. The first was amazing, and easily the best game I own. It was engaging the entire time. Now, adding extra feature and guns and plot stuff; it shall (hopefully) be epic.

It looks amazing, but I must say that the big daddy looks kinda all powerful compared to everyone else, except the big sister. He was taking guys down like they were nothing but flies, the drill even acted as a one hit kill. Anyways, I still can't wait for it to come out.

Wow! This looks awesome. I especialy liked the part where you were suddenly put under water. I always thought it was odd that the first bioshock was set intirely underwater but you were never really in it except in the very beginning.

I almost had a nerdgasm when I saw this, I LOVED Bioshock and I hope B2 has as great a storyline.
