I hear about him all the time on the internet, yet feel that asking here will be much more satisfying than using Google.

He's a marvel character who has become very popular due to being usually pretty damn funny with his various pop culture references and fourth wall breaking antic.

He is awesomesauce at fighting and can heal from virtually anything.

That's pretty much all you need to know about him.

Although, I would go to a comic shop and pick up a comic or two of his. That'd be the best way to learn about him firsthand.

He's basically Spider-Man, but with Wolverine's healing powers, lots of guns, a penchant for breaking the fourth wall, who's basically as crazy as the Joker, but only mildly as homicidal.

The funniest Marvel comic character ever. I'm assuming you don't read comics, so to help you, here's the trailer to his next game:

Wade Wilson was part of the Weapon X program, along with Wolverine and some more Marvel characters. He volunteer'd because he was hoping it would cure his cancer. He was a pretty boy, and the Weapon X "cure" brought him regeneration. He could regenerate his body as long as his brain was intact. Meaning if his head was chopped off(and it did, many times), he would grow out another body. The regeneration from Wolverine's DNA had side effects. It made his cancer grow exponentially and quickly heal from the regeneration. It spread to his brain as well, causing him to become insane and disfigured. Legions grow all around his body and head and disappear. After the Weapon X program, he woke up not knowing who he was, and escaped from W-X Wolvie style and eventually regained some of his memories. He likes money(so he can buy exploding chairs and such), so he became a mercenary. The rest of the story is hallucinations, talking with himself and being damn, damn, damn good at his job and at being funny.

If you're more interested, noted highlights are Deadpool: Secret Invasion(or anything by Joe Kelly) and any of the Deadpool and Cable comics. The D&C series is like a buddy cop comedy done right.

One of my co-workers who is a comic reader said that he once asked his adversary if they played Street Fighter, and when they said "No" he Shoryuken'd them.

I heard he also punched someone across panels at one point too.

EDIT:
first time I saw Deadpool in anything
Everything I've seen him say since I saw this was the exact same hilarity.

Deadpool while above all being funny has very serious and dark moments like the plot involving Deadpool and T-Ray who both have serious background problems. The dark moments are what i find more entertaining.
