Star Wars I would imagine.

My personal favorite is the Big Lebowski.

It widely depends on what's considered a "cult movie" and who you judge to be part of the cult, as distinct from people who just enjoyed it at the time.

I would guess Rocky Horror is up there no matter how you slice it though.

I would define a "cult classic" as a movie that is quite popular despite receiving neither commercial nor critical success upon release. Which one has the largest following?

Yeah, I was gonna go for Rocky Horror too.

I mean, Star Wars probably has the biggest following, but they aren't cult films. The Rocky Horror Picture Show is most definitely a cult film, so I think that's a more accurate answer, even though the OP didn't specify that the movie in question actually had to be 'cult'.

Umm would you include A Clockwork Orange in this? That's pretty popular.

I would have to say the Evil Dead trilogy has a good sized cult following.

I'm just going to throw in Pulp Fiction. It's a personal favorite of mine.

However, the movie with the largest fan base is probably between Star Wars and 007

The Princess Bride
Rocky Horror Motion Picture Show
The Blues Brothers

They probably don't have the largest following, but there still awesome...

Either Star Wars, Lord of the Rings, or Harry Potter.

HP's been picking up steam lately, in terms of the number of movie fans anyway.

Okay, Star Wars is not cult by any means. Nor is Harry Potter. Now Labyrinth and Princess Bride you have legit suggestions, same for Rocky Horror.

Though if we're going for cult status while being close to having a legitimately large following? Boondock Saints.

Star Wars has my vote for largest.

my personal favorites are "Ginger Snaps", "Little Shop of Horrors" and "Plan Nine from Outter Space" among many many more.
