I'm gonna have to go with the Aeon's Experimental "God Robot" from Supreme Commander 1 and 2.
That thing can destroy lesser robots by STEPPING on them. Nothing can possibly stand against that thing... well... short of an alliance between several other players.

That needs to be built, it is a must. But not in Japan.

I'm in support of the life-sized Howl's Moving Castle mentioned previously (even though that was technically based off of a western fantasy novel). The ability to go inside would be super sweet.

I would also like to mention Escaflowne. I didn't really like the show, but it could turn into a dragon.
