(A lot of this is inspired by the last episode of the Clone Wars. This info is gathered from the 6 movies and a limited knowledge of the extended universe)

Darth Sidious tells him to kill Asaaj Ventress, his second in command, even though between the two they could over power Sidious and Dooku would be the ruler of the Sith Order (all two of them). Now for any of you who don?t read Wookiepedia in your free time, the Star Wars movies as well as the Clone Wars series take place during the Rule of Two. This Rule means that the Sith Order can only exist in two people, ?a master to hold the power and an apprentice to covet it?. Now in order for the apprentice to become the master the apprentice (in this case Dooku) to kill the master (Sidious). Now, Sidious feels Asaaj is threatening this relationship and making a third Sith, so he orders Dooku to kill Asaaj. Now instead of Dooku being like ?Yo, Sidious, sorry but I gotta side with my lady friend here, so imma gonna kill you and take your place? he?s like ?Yeah dog, en bee dee.?
Now, say hypothetically Dooku had devised a plan to overthrow Sidious, he would have
A) Won the war by killing the chancellor (sidious and chancellor palpatine are the same person, just in case you didn?t catch that and if you didn?t SHAME ON YOU) which then would have allowed the Separatists to form the Galactic Empire we see in the Original Trilogy. Maybe a little more fighting would have needed but in the grand scheme of things it wouldn?t really be all that hard.
B) Become friggin? Dark Lord of The Sith. Which isn?t really saying much because after all there are only two at a time. But still, who wouldn?t want that?
And Asaaj clearly wants to be a Sith, so if she helped Dooku with her plan she would have gotten what she wanted and become the Sith Apprentice.
But no. Dooku is so in love with Sidious that he blindly follows his orders. It doesn?t help that both of them are really old, and even with the Force on their side neither of them really have much more time alive. If anything they only look a few years apart, meaning when Sidious dies of age, Dooku would be right behind him. But then Anakin/Darth Vader shows up and pretty much kills em both.
And it's not even like Dooku and Asaaj would have had a super hard time killing him. It would be an awesome fight and I'd love to watch, but Asaaj and Dooku both on several occasions hold off Obi-Wan and Anakin, who are generally thought of as some of the greatest Jedi ever. Where as Palpatine fights Yoda to a stalemate (same thing Dooku did, and Yoda trained the guy) and Vader basically picks Palpatine up and chucks him into the reactor. The only fight Palpatine is shown really winning was when Windu, Fisto and those other guys come to arrest him. Even they went to arrest him, and it wasn't until most of them were dead did Mace Windu seem to change to "Shit, I gotta kill this bastard" mode. And again, had Vader/Anakin not showed up it's debatable that Ol' Palpy would have lost that fight.
