I would take out Lucien in Fable II, killing him was not nearly satisfying enough. I think I would throw him off the top of the spire, teleport down and catch him on my sword.

And I would take out Vince from Vice City Stories, his swagga pisses me off. Really though, how long is he going to wear those military fatigues after being kicked out of the Army? Assassin's Creed has very little in terms of actual assassinations when it came down to it, I'd say the Hitman games are better for that.

it did ... it's just that it's SERIOUSLY difficult to pull off a perfect undetected assasination
