Nor has she been confirmed to be male and thus I subjectively infer Valve's hints as Pyro being a female and thus that is how I represent her.

Fang in FF13, although I guess she's not really the main tank (Snow, a guy, would be that). She's still kickass though

Screw Snow! Seriously, he's gone for most of the game and when Fang appears and acts as his stand-in, she does a better job than him. Fang is a much more appealing character, she has more attacks per hit, her counters are awesome, and she works other classes very well compared to Snow.

I honestly think that Snow was there to look cool, because I never used him unless it was absolutely story related.

Fable II's Hammer (Hanna) has been discussed already.

The only others that spring readily to mind are characters like Rainbow Mika or Tina Armstrong.
both of whom are meant to be the more slow moving and "tough" fighters in their respective games.

She can take hundreds upon hundreds of bullets because she was blessed with the magic of "uber fast healing".

you obviously haven't seen her get blown away by the shotgun alt fires of two combine prison guards during the raid on nova prospekt.
She just ran up the stairs then BLAM BLAM! body rolling back down the stairs

but yeah other then that one one incident she is pretty tank. Especially when she grabs a shotgun then NO ONE CAN EVEN TOUCH HER!

How come no one has said either Samus Aran, Terra Branford (FF6) or Celes Chere (also FF6)?

Samus is a badass wrecking machine that can take a lot of goddamn hits, Terra in esper mode will suplex Sabin suplexing a train, and Celes, a former General, can not only absorb magic attack via her Runic ability, but she's also like a Paladin class and that's BEFORE Espers come into play.

Seriously Escapists...you're dropping the morph ball here.

But yeah, she was one of my favourite characters. Her whole guardian complex thing was pretty cool. I loved how if when you get her to use the taunt ability one of her shouts is, "You come through me before anyone else!"

Bioware is generally pretty good about these. Bastila always ended up being the best close quarters fighter in my party in KOTOR, Ashley in Mass Effect has the highest pure combat stats next only to Wrex and your main party member if you wanna wreck some serious shit in Jade Empire is Chai Ka (although she is technically a genderless demon who just happens to inhabit the body of a little girl)
