Certainly one of the best in terms of raw firepower (in a handheld gun) would have to be the RY3NOv5 from Ratchet and Clank Up Your Arsenal. Or for Melee the Energy Lance used by Corvalis in Motor Mayhem.

The Rock-It Launcher from Fallout is one of the most fun weapons I know. Just ahead of the Super Sheep of Worms.
