Reaver from Fable 2 is a complete asshole. Gets his picture taken from the second coolest character in the game, learns that the picture will take 3 months to develop, and then decides to kill the photographer. WTF.

A friend of mine, who happens to turn into the biggest dick when playing anything online.
Now, I suck really badly at Halo 3, but when I had 17 kills and 0 deaths, with our team one kill away from winning, I backed off the front lines to finally get a Perfection. One second before the game ends, he kills me with a Splazer, and my other friend about to get a Perfection died from the red team(We were playing people who were reaaaaally bad..). So, he, Mr. Jerk Off gets his Perfection and laughs at us for dying. I tried to hit the boot for betrayal button, but hit the wrong key...
Of course, if you betray him before he gets a Perfection/Running Riot/whatever he'll cry for five friggin' hours.

the escapist user Abedeus. he hates WoW and Titan Quest (my favorite games).
for real, its either Bowser in mario kart wii (because of homing nuclear shell) or Biara in Heroes of Might and Magic 5. shes 1 heck of a pain.
if none of them, the extremly clever A.I in HOMM3-5 (even warcraft 3´s ai has alzheimers compared to the AI in HOMM3-5.)
and at last, the AI in L4D. because they are so damm stupid and is really slow when it comes to saving you. i would rather have human players with alzheimers than them sometimes.

Im top rank in nearly every game I own that has multiplayer, Im friendly so I try to help out people who are lowest rank and die a lot. Usually this involves covering fire, sniper support, medipacks in FPS. Most people who are new will say thanks, or just get on with the game. But some of them SCREAM down the mic, accusing me of kill stealing, distracting them.
THOSE are the biggest assholes in gaming.
Ambassador Udina from Mass Effect. Guy's a total prick, who nearly brings about the end of the galaxy (unintentionally, but still) for short-term political advantage.
I would guess that the Titans from the God of War series have some of the largest assholes out there.
Otis who will call in the middle of a boss battle and won't let you hang up.

That guy from Condemned 2. He apparently wanted to be in gears in war, got lost, ended up in Condemned, was so mad about this he decided to be the villain.
