I suppose, but you're assuming that the girl in the "trailer" is a little sister and you're also getting previous posts mixed up with what I suggested.
In my gameFAQs post (which, even though I cannot prove it, I am still gonna believe started the prequel rumours that were rolling about for a while) I said that the player should play as a secondary figure during the formation / fall of Rapture, And ultimately choose where their loyalties lie, with Ryan or with Atlas / Fontaine.
This would be cool as it would allow you to meet and interact with all those voices we were introduced to in Bioshock as well as allow for a more complicated moral choice engine to be built.
Playing as a little sister would be cool, moral choice or not, based on the fact that crawling through those pipes would allow for some interesting level design.
