I'd have to go with Samus on this one. It seems to me that, overall, every time we have a female game character she's a woman first. By which I mean the Devs make that the main highlight of her personality instead of just writing her as a believable person. This might not be a bad thing per say but it's a trend that needs to quietly die.
I'd have to say the "best tough, believeable female character" award goes to a toss-up between Zoey from Left 4 Dead and Alyx from Half-Life 2. Hm...does that mean VALVe knows how women operate?
