Cool weapon!

The paired RC-P90s from Goldeneye for the N64. 160 rounds of high-penetration mayhem with a reload time of almost nothing. Granted, you had to unlock the ALL GUNS cheat for it, but it was worth the effort.
Personally it's hard to pick one specific weapon out of the Worms series but I can say the sheep bomb, the kamikaze and the holy hand grenade stick out in my mind. Nothing is more annoying the being finished off by someone jet packing away after dropping some sheep or that perfectly aimed kamikaze that sends your last guy rocketing off the end of whats left of the level.

While not quite a video game weapon per se, my favorite would have to be the shokk gun from the Warhammer 40k Orkz.
Get this: the weapon opens a portal, one end on the weapon and the other inside the targets armor. The ork then shoves a small, vicious, and possibly rabid animal through the portal. Hilarity ensues.
This is the kind of rampant sillyness that the orkz are known for.

Personally, since I play with Tyranids, I take pride in the fact that I'm so evil that even my blood can be used to kill people (i.e Venom Cannon). But as for Video Game weapons, I'd have to go with the Super-Charged Gravity Gun from Half-Life2 and Episode 1, the Hivehand from the original Half-Life, and every physical object in Dead Rising, because of all the possible one-liners...
I reckon this list is decent:
1) AC130 in COD4 - splattering a group a bad guys with 105mm cannon is sweet
2) Turok on N64 had some ridiculous weapon with a truly massive blast radius, forget what its called though
3) Shotgun with alt fire of two rounds - very effective, Auto shotguns are great too
4) Gravity Hammer, love the sound and the effect of those little twerps flying
5) that skull driller thing from Turok again i think, lets it fly, finds the head of the bad guy and then drills a mighty hole in the back of it

One of the coolest weapon ever has to be the lightsber.
I mean come on a weapon that can cut through anything just has to be awsome.
Oooooh and the Ion cannon from C&C nothing says F.U like dropping that one on your enemy's headquarters and then invading with your 20 or so mammoth tanks.
But the ultimate weapon has to be the BFG 9000 I mean thats some serious firepower right there. loved tha thing when I first got it.

I love the harpoon gun from Crackdown. Give them a good kick and shoot. You can peg them anywhere! And then you can shoot the body in a different place so the body ragdolls differently all the way down a wall. Nifty.

The coolest weapon ever was and is the machine gun from Gears of War. The one with the Chainsaw Bayonet. Chainsaw+Machine Gun=Awesome Violent Fun
