This tutorial [http://afteractionreporter.com/2009/02/09/the-complete-and-utter-newby-tutorial-for-dwarf-fortress-part-1-wtf/] should help, it provides a save with everything needed to follow the guide.
It was a tutorial by this guy that originally helped me get into Dwarf Fortress, though that was with a different version of the game. The current series of tutorial are actually slightly outdated, but the basic game mechanics haven't changed since that version of the game so it still works perfectly as a tutorial. Just keep in mind there might be some minor differences and improvements between the version the tutorial teaches and the latest release of Dwarf Fortress.
You'll also want to have the Dwarf Fortress Wiki open in the background as well. It's extremely handy to be able to quickly look up anything you don't quite understand with a few clicks of a button.
And finally you'll also want to use a graphics pack, simply because Dwarf Fortress becomes infinitely easier to play when all the ansi symbols are replaced by adorable 8 bit art. Here's a list of mods for the game, you'll find graphics packs under said category. Personally I like Mayday's graphics pack myself, but different people have different preferences. There are also links to other tutorials and the like on that list as well. The two visualizer mods are also worth keeping an eye on, though neither work with the latest version of Dwarf Fortress yet. However once they're updated to do so, being able to get a 3D look at your fortress can be extremely handy and entertaining.

Yeah, he pretty much said everything I would have. Other than tilesets being necessary to make the game playable. I prefer the vanilla graphics, as once you learn it it makes the game a lot cleaner and simpler than the cluttered tilesets. I think Mayday in particular is an abomination. I also disagree with the idea that they somehow make the game easier. First time I played was with a tileset, and it didn't help at all.
You could also ask questions here in the user group. If you ask on the official bay 12 forums, you're guaranteed to get tons of help. They're a nice bunch over there.
Also if you need motivation, try reading some good DF Let's Plays. Best example is Boat Murdered [http://lparchive.org/LetsPlay/Boatmurdered/]. It has legendary status, and despite being from the older versions it showcases the general insanity of DF. Gemclod [http://forums.somethingawful.com/showthread.php?threadid=3349669] is a good example of the new version, though it's still a work in progress.
