That's actually a really cool idea. I would do what I did in Guitar Hero: make the cast of Metalocalypse and put them through the zombie-fest (well, almost all of them. I guess I can come up with a reason for all of them to sit out one round)
As for something original...how about Final Fantasy? If Final Fantasy I was to ever be re-made I think it would be priority 1 to allow full character customization. The original FF1 was about Fighter, W-Mage, B-Mage and, Rouge saving the world. The re-make should allow you to make, from scratch, your entire 4-party team. All female humans? Awesome! 1 human, 1 Mog and, 2 Burmecians? Groovy! 4 Moogles? The Best.
I'd also like to see a Harry Potter themed MMO in which you are your own Hogwarts student. This game would have to take place either before Harry's enrollment or, in his early years so as to keep your character from out-Harrying Harry. An MMO based around Discworld would also be great: Human, Troll, Dwarf, Wizard, Orangutan (well, maybe not that). I mention Discworld and Harry Potter because both have and, had games but you were always a well-known character(s).
Pokemon would be a good one too. Not any character customization in those except for your name...
One more: Halo. Why bother choosing that I prefer playing as a Shangheli if they never let me play as one? It's cool that I can change my armor in Reach but I want to play as anything
but a human when I'm on live. Humans, no matter how super or, mechanically/biologically enhanced, bore the crap out of me (at least they do in Sci-Fi and Fantasy settings).
