Im not sure, but timeline will probably be post GTA IV.
Judging by Samantha's description game might be set in Los Santos/San Andreas.
And also judging by Jose's description i think its not going to be all urban setting like Liberty City, witch points out a possibility for San Andreas return or a new setting.

I think it's obivous what the plot will be, you are living a crap life and get involved with gangsters or the mafia and then you will slowly walk your way to the top.

But I don't care about the plot, killing people and stealing cars ir the only thing I like doing in GTA.

Remember how little plot GTA 2 needed?
Answer public telephones and do insane missions for loonies, rednecks, hippies, mad scientists or the russian mafia.

Well on one of the Rockstar sites they dropped a hint a while back about the next game being set in San Andreas. In one of the images on the site, there was a suitcase and two tickets to San Andreas.
