I prefer to take on groups, removing members one at a time without the rest noticing, until there is one left. Then I like to spin the last one around and let them realise that everybody else is dead. It's lots of fun to do in Assassin's Creed 2.

I don't know why they continue to wear it after it starts to kill them, but thier usually dead within 15 seconds of placing it on their head

Viscerally. Either with a hatchet or ball-peen hammer. If I'm going to be assassinating someone, it'll be for something personal (not going to whore out my leet skills); so I'm going to want to watch the life leave their eyes, even if they are hanging out of broken sockets.
From half a mile away with a sniper rifle. Hitting a small target through a scope at that distance. Feels good man. It's even better when the game doesn't have AI that instantly reacts and knows where you are, so they sort of stand their trying to figure things out or dive for cover.
One of my favorite things to do in Red Dead Redemption is to sit on a hill overlooking a road at night and pick off travelers and animals with the sniper rifle. This also works atop Tenpenny Tower in Fall Out 3.

Well, if I'm going for an "assassination" then I'll either use stealth or pin the crime on an accident or someone else.
So, if I'm feeling stealthy, either a silenced firearm, probably a pistol or a sniper rifle, or just my bare hands.

Why have you been paid to eliminate the target? He is dangerous to someone in some way. Does the target know this? More then likely. Will he be protected as a result? Yes, thats why you are being hired, as they cannot do it themselves. You are not hired to serve the targets honor, or your own. You are not hired to be dramatic (unless its included in the contract). You are hired for a confirmed kill, and must execute it in a way that has you around to spend the profits... Long range, .50 solid brass BMG round from over 2000 meters away, perhaps from a M107/M82. Round will obliterate subject, near guaranteed kill wherever it impacts (centre mass) bypassing body armor. Target will be down before anyone hears the shot. Range also ensures you are outside the area of any personal protective detail. Dismantle and dispose of rifle at leisure while you drive away as emergency vehicles rush by you. Works for urban or rural settings, plus or minus the emergency vehicles

I enjoy the whole "accident" approach. One of my favorite things in a game was in Oblivion you are given the task of assassinating a guy and the best way to do it is to break into his house, find the door to a passage in the wall, go up and loosen the wall mounted animal, and then hear the "thud" as it crushes the guy while he's sitting in an armchair by his fireplace. Awesome, awesome stuff.
