I would have to say just from the release date of the games, that it would be Trials and Tribulations. Anyway, a theory as to why he uses that slang is that, because the Animus uses the user's thoughts to 'translate' the characters as effectively as possible, because the Uncle introduced himself as Mario, the Animus detected Desmond's subconsious of Super MArio- and thus put that quote in to relax him a bit.
