Depends on what you mean. The Lord of the Rings books and movies exist within their own canons.
Will this affect the official Lord of the Rings canon of the books? No. Even the movies can not change the books. Those are set in stone until Christopher Tolkien manages to somehow find enough of his father's notes to cobble together another story and that depends on whether or not you consider anything Christopher puts together from his father's notes canon. (Personally, I do.)
Will it affect the canon of the movies? Possibly, considering that both are owned by Warner Brothers. Not likely, however.
Will it affect the canon of the video games? Well, to answer that, you first must ask if the video games even have canon. They certainly draw from the movies and books (more-so from the movies than books), but do they actually have a recognized canon that they all exist within? I do not believe so, but if they do then they have been doing a piss-poor job of it.

Sounds intriguing.

I, however, will wait for the game to be finished, released and reviewed before I even think about getting excited about it.

Since they are doing this they should at least hire a good fantasy author to write the story so it stays true to Tolkien's work. Christopher Tolkien would be a good choice since he is Tolkien's son and has edited his work. Another good choice I think would be George R.R. Martin.

Dear god what a travesty. Everyone knows that the only option is to do a hungergames/middle earth crossover.

Enemy relationships are shaped by the player's actions and decisions. Enemies remember the player's previous actions and learn to adapt. They will be unique in every gameplay session. Through the system, players will have different archenemies.
Meaning if the game sees you spamming ranged attack it will throw guys with huge shields at you etc.

If they made a game that was faithful to the origial books you'd walk around the world, there would be singing and few times you'd get to turn invisible.

So it would be a hiking simulator with some karaoke/rhythm-gameplay and stealth, and while I would play it, I can't see it capturing the general audience.

So I can see why they'd take liberties with the source-material or place the game on some other area or something, but this isn't even related to the lore.

At this point I stopped to see how this can be Middle-Earth story. Lore doesn't support that sort of existence: even if Talion would die by morgul blade to be resurrected as Wraith, he would be resurrected as a slave of the blade.
I could see a storyline where he gets stabbed working; have him be infected with the Morgul-blade, and slowly turn, while trying to fight it and find a cure.
Or give him a ring or some other evil object, and have him lose more and more of himself while he uses it.
Although that would carry the risk of punishing the player for doing the 'fun' thing, and while it would work as a narrative, I'm not sure it would work in a game that is meant to have general appeal.

Lore that lends itself to a decent RPG in Middle Earth would be akin to The Children of Hurin, playing as Turin in his roles as a hunter and a leader of bandits etc would be quite neat, especially making moral choices given that Turin is destined for ill-fate whichever way he turns. That's what I thought of when first I read about this game, but seriously? "Spirit of Vengeance?"

Maybe it will be good but with your track record regarding LOTR I am not going to get excited...
