I've got a good one: The Flash.
Make it a sandbox game, similar to Arkham City. Flash has his own movement system already built into his character (running fast, running up the side of buildings, running fast to make long jumps, and running over water), so that's one problem down.
Flash has a rogues gallery almost on par with Batman, so there's no need to worry about a shortage of characters.
The main villain? That's easy: Gorilla Grodd. Plenty of other villains to work under him: The Shade, Mirror Master, Captain Boomerang, Weather Wizard, Captain Cold, the list goes on.
The building blocks are there for a really great superhero game.
