I am going with D. Not only is he a badass, but they actually make his struggle to overcome his vampire blood an actual stuggle. Blade and Alucard dont seem to have any real side effects or urges to overcome, just faux emotionlessness and angst (respectively).
Plus, D doesnt just have to deal with vampires and werewolves, he has to deal with beings of various different fantastical and frightening powers. He has to be seriously cunning and versitile. I would say the same for the Belmonts, but frankly they dont really have foes quite on par as Reiganse or Benge as far as unreasonable abilities.
As for Van Helsing...I didnt like the modern adaptation, it was terrible. Two lines that had me laughing like an asshole in theaters were "YOU TOOK MY FINGER!" to an obviously folded down finger when he had all 5 fingers on both hands for the whole movie until then, and "I FEEL NOTHING!!" delivered like a 12yr old yelling at his dad that he isnt a child when he is supposed to be talking about being totally devoid of emotion. It may be a bit unfair of me to ding his awesome based on the movie he was in, but a good hero can save a bad movie...he didnt sell it to me. Basically it's steam-punk setting Batman if Batman was far less intelligent and less competant.
Buffy is pretty much a pop-culture joke (Admittedly I LOVED the movie...not so much a fan of the TV show). I will keep objective and not go into other stuff I dont like about Buffy. She has superpowers, but no grasp of strategy. Her villians can be varied and interesting, but will all the help she gets from supporting characters there is no shortage of someone knowing/finding the weakness so she is hardly capable as a stand alone (not that you always have to be alone...but you shouldnt need an entourage everywhere you go. Also she is mentally unstable.
Lastly, Vampires in D's world are serious badasses, only Dracula from Castlevania can come close. The DEMONS from buffy arent even that scarry. It takes one hell of a badass to take one down. My vote goes to D.
