She is probably my favorite female game character and her breasts are not her selling point, in fact I spend more time watching her face (great facial animation tech Valve!) than the area below that.
As long as certain parts female's *ahem* anatomy *ahem* are not used as one of the main selling points of the game, I see nothing wrong with having the women having bigger breasts.

1) D-cups are ridiculous. Never break C unless it's a defining character feature.
2) Eyes. Eyes are the gateway to the soul. A lot of anime characters talk about "killing intent" and the "look." If you look into a rape victim's eyes, they usually have a "hunted" or "haunted" flicker in the deep part of the eye. It's hard to describe, but if you give her large, expressive eyes and possibly a tragedy or character flaw from the past and successfully translate that into the eye, she gains depth.

Well a good example of all of that would be Mona Sax. Money being the flaw.
She is a good example having a little sex. Thinking at first she is just a hot toffee then pulls out a gun proving she's a dame. Now a dame is what I personally expect, personality and body are top of the line with a weapon to kill with. Though she is not as daunting as the villain. Nicole Horne proves that the best villains are a *****.

Hmm I dunno; my wife's got DD's that weigh almost a kilogram each and even with a sports bra she can't run full tilt without doing some damage. Out of the six years I've known her I've rarely seen her even jog.

When I'm doing non-sport jujitsu, or Kendo, sparring or any other of the activities I do I have to, and I mean HAVE TO strap mine down, as my motion is actually limited by them. and yes the strain on the back and shoulders is horrid.. only real way to combat that is reduction, or the path I take which is heavy weight training on the major muscle groups you use to support that weight.
and for the love of....the only thing that really bothers me, is the games like DOA and such that show they know NOTHING of breast physics.

B: They do not defy the laws of physics by one breast swinging up, and another down...they stay together.
C: if they are real...they will NOT stay nice and shapely when the character is laying on the ground.

in reality though I don't see nearly as many super busty characters as one thinks...yeah, massive milk wagons may be more memorable, but when looking at most female characters be them PC/NPC, Main/Support characters...they tend to fall into the large B to mid C cup range...which is actually pretty average.

Survival horror?) where a character DOES have big breasts, but is also honestly overweight, maybe a little clumsy(I mean CLUMSY, Not "Beutiful Damsel in Distress", Stephenie), and actually HAS some problams with acting athletically?

This topic fails if you havn't played the game where the character is in because then you are just comparing pictures - where looks are everything (Ivy wins).
However the personality of a character can change your outlook on that person, they maybe less physically attractive but their character is more of a turn on (as long as they are not too ugly).

the girls from mass effect, faith from mirrors edge, velvet assassin(i think i duno i aint played it) the perfect dark woman hasn't got big ones, but she wears a tight suit..so same thing i guess.
