Secret of Monkey Island, Day of the Tentacle and Earthworm Jim all leap to mind for laugh out loud and off the wall humour.

Portal 2 for intelligent jokes.

And Dragon Age: Origins for snarkiness, sarcasm and Ohgren.

Champions of Norrath: Return to Arms
I loved, just loved playing it with my big brothers and one of their friends, it was so much fun, I'm sad that games like that have started to die out, such bullshit that modern games have to have only online co-op, i want offline 4-player co-op, that's the best kind of co-op. Oh such good memories about that game, no game since then has really lived up to that, maybe Oblivion, Skyrim or perhaps DA:Origins

Am I the only one annoyed by "what's your favourite (Insert media here)"?
I find it nearly impossible to pick a favorite game, as do a fair few others here. but anyway...

Timesplitters 2 and 3, one had some funny character select but lack cutscenes and the like makes it look a lot less amusing than TS2 or 3.

Thankfully someone mentioned Leisure suit larry. Was the first game I thought of when seeing the thread title.
Loved Roger Wilco's adventures too in space quest 1 and 2, had plenty of laughs with these!!

Blergh. It's been years, but I swear the words "Your creatures are fighting amongst themselves!" will still send me into a frantic clicking frenzy to try and stop the trouble.

I'm quite surprised that nobody has mentioned the Penny Arcade: On The Rain-Slick Precipice Of Darkness series yet. It absolutely cracks me up, it's like you're actually playing one of the webcomics. I love those dudes' sense of humour though, so it really is tailor-made for me.

It's so funny I'm even struggling through the third episode because despite the gameplay being much weaker and the 8bit style/music getting right on my tits, it's still hilarious.

Don't play many games I'd consider funny so this shouldn't be hard.

Borderlands
Saints Row 2
Bulletstorm (This wore off after a while because it just became repetitive)

Yakuza 4 made me laugh out loud the other night (I'm new to the series). I was playing ping pong against a female patron of the bath house, and the whole silliness of the situation hit me and I couldn't stop cracking up. This game is called Yakuza, and I've just smashed in the heads of 20 random angry people on the street. Now, I'm playing ping pong in a bath house, and I get bonuses for sneaking peeks at her. The juxtaposition struck me as hilarious and I laughed out loud. I was giggling very hard for a good few minutes. The overdone animations and the smash hits that caused the female patron to fall over and swoon for you were also ludicrously funny at the time. Being really tired made it much funnier as well, I'm sure.

I'm really enjoying Y4; feels so much like Shenmue. Or at least it feels like my fragmented and foggy memories of Shenmue. So long ago... all I remember for certain is I always blew all my money on those collectable figurines from the "junk machines" in the convenience stores.

Just to be spiteful, I NEVER laughed out loud at Saint's Row 3 :p. I did enjoy the game; I beat it and all the DLC I got for free with it (about a month after it came out, THQ's site had a sale with the DLC Season Pass included).

Mario Party 6.

On a similar note, the same game, and I dare say, the same match of Mario Party I'm talking about, is the single most blood boiling game I've every played.

Borderlands or any of the Ratchet and Clank games are full of funny dialogue and weird characters.

Am I the only one who finds Skyrim funny? A bit of the dialog and situations are pretty funny. Especially a bunch of the Daedra quests.

Super Mario RPG: Legend of the Seven Stars es #1. Seriously, I love the innocent humor in it. Bowser and the Princess are a riot.

Magicka.
Not only does it supply it's own humor, with an ungodly amount of references to pop-culture and down-right ridiculous scenes, but killing each other is funny as hell.
This, Magicka is great, I've had to pause that game to wait till I stopped laughing because I couldn't see through the tears!
