I don't know, and I don't want to know. From what little I've seen it looks damn stupid.
Plus the fans seem crazier than bronies... Just not as obvious as there's not as many of them in comparison to bronies.

In short, it's really weird in kinda of an awesome way. The only way for you to figured out what exactly homestuck is about is to read it for yourself, though the first act is a tad long and really tedious at points. So if you do give it a shot be prepared for a bit of a slog in the beginning.
A long web comic with a really incomprehensible plot with really annoying/batshit insane fans on tumblr. Hey, capcha agrees with me ("words of wisdom").

Hey, Homestuck fan here. It's a multimedia webcomic.

The video above may give the misleading impression that the comic is nothing but flash animations. It actually currently has more words than the Holy Bible.
As many people have already pointed out, some of the fandom is pretty rabid. It's pretty easy to ignore them.

A really long webcomic with the occasional flash animation, dialogue heavy logs of text that detail the main characters interaction, interesting plot, assuming you're willing to read through it a few times to understand it, the characters are decent, though take a while to flesh out fully, and it's timeline is incomprehinsible at times.
It's mind-blowingly awsome, yet tedious at points, but is, all-in-all, a decent webcomic with interesting characters, an interesting world, and an interesting story.
