Well the the dialogue in the original Resident evil is hilarious in an unintentional kind of way, "you were almost a Jill sandwich", cracks me up every time.

I found Amped 3 to be pretty funny.

The Paper Mario series (well mostly Thousand Year Door) was funny in it's own juvenile way.

Portal was pretty damned funny, don't know, games I've played don't tend to humor.

I found Giants: Citizen Kabuto to be hilarious. Kinda like Monty Python in a video game.

Peasant's Quest. its a flash game on Home Star Runner. i find the text based options that you can do are hilarious.
"put baby into bucket"
"Okay Father of the year.. You put the baby in a bucket and it quickly descends deep into the well. Yours is a black, black heart."

1. Psychonauts
2. Ratchet and Clank: Up Your Arsenal (the best and funniest of the "Ratchet and Clank" series)

Portal, Curse of Monkey Island, Earthbound

Saints Row 2 - It just has sh!tloads of funny ass jokes and hilarious things you can do like skydiving naked and blowing up everything in cheat mode.

Portal and Psychonauts. Not coincidentally, both written by Erik Wolpaw. Portal has GLaDOS's snarky dialogue, and Psychonauts has two dozen deeply fleshed-out and well-defined characters, each with their own bizarre qualities. In a lot of games, you talk to everyone to look for someone with valuable information; in Psychonauts, you talk to everyone because they're all fricking hilarious.

About Psychonauts.

Exactly my point. It had a LOT of serious overtones to it as well, but BOY, it never missed a beat with its humor. Example (one of many) Raz's training with Sascha is incredibly hilarious.

EAT LEAD! The Return Of Matt Hazzard, was/is (still in progress) incredibly funny. Will Arnett and Neil P. Harris are HILARIOUS.

I guess golden axe, because I had so much fun with the co-op.

The Monkey Island series! Going from the weird pirate that is advertising another Lucas Art game, to your sword skills can only be matched by my wit! and of course you burying the Evil Ghost Zombie Pirate Lechuch with tons of ice under his own amusement park.(Yes evil pirates make theme parks.)

Also Lechuck is the most badass villian ever! He parks his boat in rivers of lava. LAVA!
