It's from Once Upon a Time in the West (another one of Sergio Leone's spaghetti western). It's one of the best scenes of all time.
I feel compeled to add that Sergio Leone's westerns are some of the best, IMHO.
They definitely are. Everyone mentions The Good, the Bad and the Ugly as their favorite ending duel. I still think For a Few Dollars More is the best final duel of them all:

In the end, Ennio Morricone makes Leone's films legendary.
My top three favorite western movies is the whole "Man with no name" trilogy. I think my favorite is possibly A Fist Full of Dollars.
Do yourself a favor and check out Kurosawa's Yojimbo.
I'd second this. I watched Yojimbo, Seven Samurai, and Sanjuro recently having been given a box set as a gift.

I was enthralled, they managed to hold up to modern standards without the aid of nostalgia. Kurosawa and Mifune's partnership was genius.
