Probably Mass Effect 2. And now that we all know Mass Effect 3 exists, it's time for me to start spazzing about that.
Two things:
1. This. ME2 had me really hyped. Went to the Midnight release and kept tabs on any info I could find on the 'net.
2. Bub yer avatar is really hard to look away from. I've seen it many times over the months, but finally have to say something since I quoted you anyway.

I think I was most excited for FFXIII since that was the first (and probably only) time I stood outside at midnight to get a game.

New Vegas is the one I'm really waiting for. Hope they do it justice and I suspect they should be able to.

Thanks to Blizzard my decade hype for StarCraft 2 just died along with Diablo 3. If I played Mass Effect 1 before the release of Mass Effect 2 then ME2 would have been the most anticipated title for me. Right now I think the most anticipated release for people overall is the new Halo for 360 owners. The first release of the announcement for Halo:Reach had overwhelming support. As an fps player, I also do think Halo: Reach. Most anticipated DLC however, is the 4th DLC for borderlands (who knows which gun company is evil? Tediore or Dahl??)

I was most hyped for Persona 4. I hardly slept the night before and almost annihilated my local GameCrazy clerk for refusing to give me an artbook with my pre-order. I got it in the end.

Right now the game I'm most excited for is Blazblue: Continuum Shift. I can't wait to play what's sure to be the greatest fighting game ever made. July 27th can't come soon enough.

I will never get hyped over a game release ever again after I got so excited for sonic heroes... But yeah world wide starcraft 2 probably has to be one of the most anticipated games ever.

Probably Gears of War 3. I'm not that excited right now, but it'll probably have "the christmas effect".
About two or three weeks before it comes out I'll go from, "Oh yeah, I forgot about that" to "OHMYGODIT'SGEARSANDIT'SNEWANDIWANTITNOWLKJGVIBK!".

The game which I remember being the most hyped for was GTA: Vice City - I remember constantly looking up new information about the game, and I pre-ordered it almost 3 months in advance. I did something similar with GTA: San Andreas. Both games were fucking awesome.

For me, it's The Last Guardian. Gonna be the GOTD (Game of the Decade) for me. Shadow of the Colossus is my favorite game ever.

Whatever Respawn is making. I wanna see what they can do with no strings attached.

My most anticipated game was probably either Modern Warfare 2 or Alone in the Dark 5. Believe it or not, I was only dissapointed with Alone in the Dark [sub] Which Alan Wake stole many of their ideas.[/sub]

Gears of War 3
Mafia 2
Mass Effect 3
The Elder Scroll's V: [Something]

Just recently:
Red Dead Redemption, it blew me away... but now I've realized it's just like every other rockstar game. It's cool for a week or two, but then you realize there's nothing left to do.
I was also pretty hyped on Mass Effect 2, I had to calm myself down by playing through Mass Effect 1 a few times x)

AAARGH! So much hype!!!

San Andreas was indeed LEGENDARY! Just a shame they lost the touch with GTA IV : / It got too realistic and serious. Also, there was no landscape... only a damn city!!! What were you thinking Rockstar!?
