Metal Gear Solid 3, tossing out my rotten food during the fight with The Fear. I was very surprised to see him gobble up my discarded junk and essentially kill himself with a belly ache.

Considering those bosses are always undead, it's more a matter of intuition and at this point it's a sort of in-joke for fans of teh genre. The only time it was made apparent to me was fighting the salt lake boss in FFVIII, and that's the second or third undead boss the game throws at you.

Getting raped by Psycho Mantis repeatedly until my cousin comes in and tells me to switch controller ports. "That's the stupidest thing I've ever heard," I say. Another few times getting pwned and I try it. "No freakin' way!"

Also, similarly:

Assassinating an unarmed The End when you first see him, as he frantically tries to wheel himself away. Cruel, funny, unexpected. Couldn't believe it worked.

Final fantasy 7:

the leader of the GI tribe in cosmo canyon
One of my party died so i fished through my inventory super quick before the enemy attacked while i was scrolling
but in my haste i overshot and went past the fallen member and used a phoenix down on the enemy
which killed it for some reason

.... oh did i just win?

i've since learned that the Final Fantasy series loves to make zombie enemies weak to healing magic all the time

Baldur's Gate 2, there was an optional Dragon you could fight, Firkrag, who dropped the best sword in the game, The Holy Avenger. I couldn't beat him with my party, and I didn't want to leave the guy unharmed with me, no matter how underpowered I was. Was playing a mage, ran out of all spells other then "Flesh To Stone". Reminds me of IceWind Dale. Get teleported to the tropical region that you have to fight through all the Yuan-ti, and then the exit portal is guarded by The Guardian. Caps for emphasis.

Got an old one from Gears of War. I was playing on Insane by myself and just couldn't close out the deal and kill him. Eventually I had to go to work so I just charged him (so that I would die faster) and randomly decided to stick a grenade on him. Turns out that was exactly when his shield went down and I ended up beating him

Remember the first LotR game? There was this annoying boss at the end of like the second level (bear in mind my brother and I were young lads back then..), it was this bashing tree thing. Very annoying. We died like 2000 times, no kidding, and then finally gave in - we called dad to see if he could find out what to do. We had these crappy mouses with the little balls in them, which you had to clean every once in a while. It hadn't happened, and annoyed the crap out of my dad when he tried to fight it. We found a safe spot already, and all we could do was throw rocks and melee attack. As he was cleaning the mouse, he accidentally clicked and whacked the tree's 'arm' at just the right moment... BOOM! Boss dead.

Yeah... basically, that.

Also, I had this one epic bayonet fight in medal of honour: rising sun once... There were like 100s of the guys, I stabbed them all, and then a friend of mine was like: DUDE! That was awesome!! And then ONE samurai guy killed me. ._." All because I decided that I would kill him the fancy way xD

Absolutely Duke Nukem: Manhattan Project. For sure, no doubts.
Mine is the super fembot boss in the penultimate chapter. I got there with a fair amout of health and ammo, yet I couldn't get it right. I kept jumping over her, only to freeze in the air and fall on her lightning whip. I found a little tactic that at the begginning she's slower on the attack, so I could waste down some of her health, but then she'd always somehow push me off the platform or beat me to death. Then, one time, I just sat there and fired, no stopping. The funny part: she didn't do anything. I sat there just laying down machine gun fire, and she did nothing. I didn't lose any health at all. It was hilarious.

Mine is from a game called Rune Factory 3, an awesome game that has totally slipped under the radar because of its mediocre predecessors. (Go play it. NOW.)

I was fighting the "+" form of a boss named Death Wall, which had separated his head from his titular wall and started floating around, attacking me by changing forms and firing lasers out of his eyes. I failed this fight several times, despite how overpowered my character was, because his "drill" form was way too fast to dodge, and took most of my life bar if I got caught in it, if it didn't outright kill me.

My eventual solution? Run right up to him when he gets into drill form. When I got right next to him instead of running away, the AI had a brain aneurysm, causing him to travel around me in circles instead of targeting me. Since my most powerful attack was fire-element (strong against his ice) and deals upwards of 10000 damage at extremely close range without a damage multiplier, he was in the ultimate vulnerable situation when he was supposed to be using his ultimate attack.

Not really a boss fight, but I remember playing a game of Counter Strike a few years back. I was crouched, waiting for the last person on the enemy team to come get the hostages when all the sudden my arm seizes up or I stretch or something, causing me to accidentally hit the mouse button. Headshot, through the wall with my Deagle.

I left the game soon after that, because there was no way I was going to be able to talk my way out of people thinking I was a wall hacker.

I caught Mew using only a single pokeball.
This was back in the day when pokemon was black and white.
I smiled all week.

Edit: Err, that is black and white as in the colours in the game, not the recent Gen V versions of the games.

I was playing Crystal Chronicles (gamecube) with my younger brother (I've played it severeal times, so I know how to kill most of the bosses). I play a clavat (human, high defense fighter guy) and he plays a Yuke (weird armor-like mage guy, can turn invisible if they hold the "defend" command)

We went up against the orc king (boss for the mines). We bring his health to 1/4, triggering his self-destruct-charge-up-move (you need to kill him before he fully charges it). Due to the fact that my bro spent 10 seconds mashing the attack button when he was set on magic (which needs to be charged to use it, not mashed!), and then panicking, the boss one-hit-kills us.

The second time around, the boss triggers the attack again. I tell my brother "Ok, THIS time, we don't panic, we just mash attack and kill him before he kills us!". ...My brother doesn't listen and panics, deciding to instead turn invisible and freak out. I find myself being the only one hitting the boss, and then...*MASSIVE KABOOOOOM WITH SCREEN ZOOM!!!!*. I turn to my brother and say "DAMMIT, I told you to attack! The only way to kill that boss is to kil him before he-". And then I blink in surprise as the boss teeters over and dies, and we go to the results screen.

....Yeah, the invisibility made my brother invulnerable to the one hit kill full-screen explosion. I was shocked.

The one thing that comes to mind is KotOR. I remember having a lot of trouble with Calo Nord for some reason, so eventually I just set about 50 stacked mines in front of him and ran like hell. He died a horrible death. It was funny.
