On the planet Zork during the great war of 6,666 when satan rose from mars to attack the japanese cat people (who live on zork) however the cat people can only be killed by one thing; heavy music, so the devil rocks out a heavy rock solo on his demon guitar.
also there's booze and naked cat-girls. . .

Pre-colonist North America: that would be kind of badass to play as a member of any number of American Indian tribes, and maybe have to fight the spirits of their lore.
And it's not like they never fought each other. That would be epic as hell, if the tactics they used against settlers were the same that they used against each other (stealth, ambush, raiding, guerilla stuff generally) then can you imagine a greater game of cat and mouse-with-tomahawk?

You might have to dig a bit more to get the context for that, but I know that in what would become French Canada the Iroquois Confederacy was very unpopular among people who weren't Iroquois because the had a tendancy to go on the offensive. The Cree, at least, were very happy to have the French settlers help them deal with their Iroquois problem. That's part of the reason Benjamin Franklin had some contact with them, the French and their allies had driven them south and they had allied with the British. Or just take the gameplay I sort of described and apply it the the spirit thing. My premise would be good if they ever wanted to go WaW on it.
Both ideas would be very interesting to see. After all, Native Americans don't get explored very often in most mediums, let alone games, unless you count westerns, where they are more often depicted as a blob of baddies or as spiritual people fighting to keep their sacred land.

But now I'm giving you a chance to spoil my attempts at finding one:

How about: A Night Club, Heaven, 10th Century China, Celtic Mythology or a bar?
While I am hardly very knowledgable about settings, I think Heavy Rain covers a Night Club, and GTA probably covers a bar at some point.

I got it, Space Marines, fighting in some mystical land where magick is bringing on a new industrial revolution, and you control the game via a guitar peripheral.

Or you play as a time-travelling dinosaur who must travel through time to do the taxes of important figures in history.

How about the inside of a body?

Like you travel through bloodstreams and battle in the Stomach.

That or Neptune.
I believe there is an educational game that was developed to help kids fight cancer and diseases by the power of placebo. They controlled a white cell (which actually was a well-designed character) shooting bacteria and viruses.
Damn really?

Well I'm still sticking to Neptune. In-fact the rest of the Gas Giants as well.
If no one has told you yet the gas planets have been done in Lenny Loosejocks

Has there ever been a game set during the Falklands War?
I remember playing a strategy game on the commodore 64, it was long time ago and i dont remember the name. Also there was going to be release an argentinian game about that war, i hadread about in a magazine, never saw it, beside it was programed for a computer that i thnik never make it to the market.

Back to topic, there are many locations that havent been used for video games, for example Pergamino, Buenos Aires, Argentina. Some i guess will never seen them cause they are too plane, with nothing special that can set them apart from other place.

Ps.: correction, the game was Falklands 82. http://www.thehouseofgames.net/index.php?t=10&id=268

I dunno if it's been said or not, but how about a Post Post-Apocalyptic setting? Civilization is back on the rise, nuclear fallout still a problem, just not as much. Or however someone else thinks about it. I think it would be an interesting setting, sort of like a futuristic wild west in my opinion.

I like the idea of a true stone age RPG.

Of course, the SETTING has been DONE in the first Age of Empires game, but that was an RTS. For that matter, Civilization also had a stone age period.
