Probably the original Final Fantasy game, it has an excellent combat, a jaw dropping story, and a gigantic world (for that time). Honorable mention goes to Half-Life.
Actually, Final Fantasy was a blatant rip-off of Dragon Warrior.

I have no idea if anyone has said it yet, but...Shadow of Colossus gets my vote. I've never played any game like it.

Correct me if i'm wrong, but i found Bioshock to be quite original... The idea of this underwater city for the brilliant minds of the world. The story line was uncommon for its genre and some of its features i had never seen before in any previous game.
You have heard of Atlantis right?
Oh and...hmm... I don't know what is the "most original". I think SC brought a massive amount of important changes to the RTS genre...

Most original as in never done before in any form of entertainment? Out of the ones i played, i'd go with Project Nomads and Steamland.

I would say serious sam. There was no other fps like it and it made me very happy when I played it all the time. It was even more fun to buddy up and just go on a killing spree! I wish they would make more fps's like it, the closest that i know of is L4D but other than that i have no idea.

I've got to go with Portal (2nd favorite game of all time) or Pikmin. Honorable mentions to Killer 7 and No More Heroes. Suda 51 is officially insane...and I love it!

"Most Original" is a bit of a misnomer. If something's original, then something else can't be more original, it can only be original in its own way. I mean, Pong was original, and if you wanted to create enough of an argument, you can say nothing else is original because Pong used the video game idea first.
Sorry, I'm being nit-picky.

Anyway.

Given to answering what I believe as one of the best original concepts, it would be Metroid Prime. The ideas in it were all used before quite well, but having Metroid from a first-person perspective ended up being my favourite of all Gamecube games. This is mainly because no other shooter I've played gave me the rich lore of the Chozo or the ample information on the Sheegoths of the Phendrana Drifts. I never felt like the lore was replicated as well in the sequels, which were nevertheless very good.

Ironic considering it was a sequel, but I'd have to say Grand Theft Auto III. The first two games were way too limited by their perspective and didn't really seem to know what to do with themselves, but once 3 came out it completely re-defined the sandbox genre and for many was their first real experience with "doesn't matter if you've beaten the game to death, you can still load this up and play it however the hell you want."
