"Most Original" is a bit of a misnomer. If something's original, then something else can't be more original, it can only be original in its own way. I mean, Pong was original, and if you wanted to create enough of an argument, you can say nothing else is original because Pong used the video game idea first.
Sorry, I'm being nit-picky.

Anyway.

Given to answering what I believe as one of the best original concepts, it would be Metroid Prime. The ideas in it were all used before quite well, but having Metroid from a first-person perspective ended up being my favourite of all Gamecube games. This is mainly because no other shooter I've played gave me the rich lore of the Chozo or the ample information on the Sheegoths of the Phendrana Drifts. I never felt like the lore was replicated as well in the sequels, which were nevertheless very good.

Ironic considering it was a sequel, but I'd have to say Grand Theft Auto III. The first two games were way too limited by their perspective and didn't really seem to know what to do with themselves, but once 3 came out it completely re-defined the sandbox genre and for many was their first real experience with "doesn't matter if you've beaten the game to death, you can still load this up and play it however the hell you want."

One of the most original games I've played in recent years is Farenheit.
Yes, the story gets quite silly, and the ending leaves much to be desired, but the fast-paced decision making and gripping narrative had me hooked all the way trough.
It was fast paced, yet deep and immersive, and even though it left me facepalming myself silly during the third act, I still had to finish it.

Honourable mentions go to Portal, and to some extent Mirrors Edge - for turning the standard FPS on its head and trying something new (IE: removing the "S" part of First Person Shooter)
