Alma in Ninja Gaiden. the first time.

Ouch
Yep. I can beat her decently on Normal now, but Hard and Master Ninja are still insane. Damned grab attacks. Stupid dodging. And she never touches the ground.
Flying swallow tends to do the trick, even if it's a bit cheap. if she swoops at you jump and Y her in the face, then she drops to the floor for like five seconds, giving you a chance for a few speedy attacks. It's not perfect, but I've beaten her on Hard once. Unfortunately i died like right after. Never got over the pain...
Not foolproof enough, sadly. She dodges it and hits back way too many times for it to be effective.

Damn near EVERY enemy in Gothic 3.

Seriously, I dunno what it is, I'm playing on Normal difficulty and dying in four(!) hits. That game's brutal. Especially since attacks stun you momentarily, and a lot of monsters attack really fast.

"robert de sable" in assasin's creed, the first few times i tried to fight right there by the stage, i kept getting raped by the archers until i thought of running away in to an alley to fight. then i kicked total ass because combat is easy to master in that game.

First run through of Bloodlines, Ming-Xiao. My character wasn't much of a fighting machine and when she went into creepy tentacle-sludge monster mode, I was stuck for days. Eventually I had to use a bug to sneak behind her and unload all of my weapons into her back... I still had to resort to using a sledgehammer to bring down the last of her health though.

All of the one's in Dead Rising.
Killing zombies is fun- Attempting to dent the health bar of some over-powered ***** on a motor-cycle isn't.
Also everyone who uses commando on MW2 feels like a boss to me.

The marines in the first Half Life on hard mode, they will remind you over and over how silly the idea of a research physicist taking on a special task force can really be...0o

the 2nd el gigante in the trench in RE4 gamecube i f***ing gave up and went the other direction because its impossible to move let alone hurt him i warn anyone who fights him
Lure him underneath the boulder and shoot it, it'll fall and crush him for massive damage. It won't kill him on professional, but if I remember correctly on normal it takes off so much of his health he might as well be dead.

As for me, Demon's Souls is the current flavor of my pain, but I'd also have to give mention to the cannon sequence in Dead Space. Not the one where you kill the boss, just the asteroid shooting gallery. I bet I did that slagging thing fifty times.

Elizabeth Greene in prototype. she has a main health bar, that regenerates, along with three smaller health bars you have to destroy before you can even touch her. toppled with tanks attacking you and massive hunters jumping everywhere. and the three smaller health bars regenerate. and it fires rocks and green plasma at you that magically lock on to you.

yeah it's a ***** to fight.
She was a ***** but I never actually got killed by her in my three playthroughs (I admit I was on easy), it just took me 40 minutes, take down health, she uses ultra attacks, hide in a street and regain health by killing Hunters.

As for me, that thing that comes out of the wall in Devil May Cry 2, I still haven't beaten it to this day.
I defeated her by climbing on top of a skyscraper next to her while she was underground and when she popped up I jumped down and did one of those devastator or whateveryoucallit moves where spikes come up from the ground. The higher the fall the higher the damage :)

Another hard boss that pops into my mind was some sort of cold demon in Doablo II, she ( at least I think it was female ) could barely be called a mini-boss, but she used ice arrows and froze me solid in 2 or 3 shots, it was barely doable with a Sorceress or an Amazon, practically impossible with a Barbarian and a Paladin. Christ, even Andariel wasn't that diffucult :)

Nearly everything in Bayonetta, on normal mode, is bloody hard. Mostly because I can't time my dodges, but also sort of because I'm like

JAWSUS WHAT THE HECK IS THAT WHY IS IT EATING ITSELF OH GOD THIS IS LIKE MY NIGHTMARES OH GOD IT'S ATTACKING WHAT DO I DO.
You just convinced me to buy that game because that is the most insane screenshot I've ever seen in my life.

For me, Ultros.

I hate, hate, hate, HATE, hate hate HATE hate hate HAAATEEE HAAAAATEEEE Ultros! Mostly because he shouldn't be as hard as he is but those stupid rats wore me down.

Read through every page to make sure I was first to throw in The San-Bone Trio on Legendary mode from Gitaroo Man. Possibly because I'm the only person who's played it because I refuse to admit anybody thought it was easy because it is, in actual fact, impossible.

I'd also like to throw in the rotating blade walls in Hades from the first God of War. Not a boss or even an enemy, but they were the only thing that ever came close to making me quit an otherwise fun game outright.

First time you fight Virgil at the top of the tower in Devil May Cry 3, that is probly the most I've EVER raged at a game. (Moth boss whos name I cannot recall do to mental blockage to prevent mass killing from Link to the past darkworld)

The amount of people bringing up Rpgs and even Final Fantasy bosses mostly is odd to me, lots of you people should go try some SMT games =S

]Fucking SEPHIROTH in Kingdom Hearts Two. 13 health bars, NO ALLIES [they disable Donald and Goofy], one-hit kill moves that can only be blocked with reaction commands, and when you get him down to the third bar, he fucking RAINS METEORS that if the even touch you, YOU DIE.

I've beaten the game thrice, i'm like level 87, and i have the goddamn Ultima Weapon keyblade and i still can't fucking get close...

Lol really? I beat him on my first playthrough at level 68...this was on normal, of course, so maybe we were playing 2 different difficulties. I won't lie though, it did take me quite a few tries, and I originally tried when I was around level 55.

OT: Though it's already been mentioned, I'll have to go with Dark Link, in Ocarina of Time. I've managed to kill all other bosses on the first go, he's the only one that makes me try several times.
