Yep, on multiple occasions in both games and movies/tv. A brief list of things that made me cry to some degree or other, either tears of joy or sadness. I can't say I've ever been blubbering and lip flapping crying, but I've definitely had tears running down my face that I had to wipe away to see.

Oddworld: Abe's Oddysey : The good ending, the imagery of how the other mudoken band together to save their saviour, and then cheer when he is rescued and returned to them, love it.
Samurai Jack: Season 1, Episode 10: Jacks vs the Lava Monster: The motivation of the Lava Monster, and what his plan was, and how he kept defiant to the end. How in his defeat, he truly won, and shouted his victory and defiance at Aku. His building roar as he changed back to his true form, and then how his faith was upheld in the end, giving him his rest. Just...fucking...amazing. The voice actor that did his performance was fucking spot on with the emotion of it.
Homeworld: Cataclysm: Again, the ending. The still frame epilogue, explaining how things panned out. The final shot of the Kuun-Lan, flying in orbit towards Higarra, with all the ships of the other Kith arrayed like an honor guard before them, the narrators deep voice resonating with emotion. Just, great.
inFamous 2: The good ending. It was just well done, since this game is more recent I'll not spoil it by saying why exactly, but it was a very emotional ending, and very well delivered.
Let's see.....can't think of any others, but I know they exist. It's usually a response to something I find really awesome, not so much from sadness, though that does happen from time to time. But yeah, usually tears of joy/awesomeness at a story well told, with a very satisfying resolution in some way.
Oh! Just remembered a new one. That Futurama episode with the dog. Holy shit that was so fucking sad.
