GlaDOS, though instead of a letter it would probably be a 32 or 64 or 128 or 256 or 512 or 1024 or 2048 or (well, I think you get it) bits data-package.

Uhh, I can't decide.
On one hand: Kerrigan, hive-mind and leader of what is arguably the most powerful and versatile force in the galaxy. And yet the loneliest woman in the universe. She could clearly use some comfort (as long as she doesn't stab me with those long arm/wings of hers).
On the other hand: GladOS, that poor, poor little AI. All she needs is a comforting hug and to not open the vents to the nerve gas as we do science together.
On the yet another hand: Harley Quinn is hot. She might be quite insane, but hooo-boy. Look at those melons.

This question reminds me of how few memorable female villains there are in games.

OT: Neither of them. Frankly, the only evil female villain I can think of is Sylvanas. But she's dead, and I'm not into necrophilia, so... Wouldn't work out.

Kreia is actually Arren Kae, Brianna the Handmaiden's mother. Think about it, white hair, similar intentions, and Kreia gets a lot more defensive whenever Handmaiden is on the ship

Or from games rather than comics, Killer Bee Cammy and the rest of M. Bison's Doll Agents are all very much in my affections. I love those odd outfits (the mini-ties!).
