Duke Nukem 3D was the first thing I did.
I was 3 years old- luckily most things went right over my head, but it was fun to shoot the crap out of baddies.

Hm hm... i remember having a few smaller games before playing "LHX: Attack Chopper" (only game i ever knew where you could lock a heat-seeking missile onto a camel and blow it up *g*) and "Stunts" in the early nineties.

But i remember totally getting hooked and playing a LOT of the golden classics. The Lucasarts games, X-Com: Enemy Unknown (Still one of my favourite games EVER), SimCity, Prince of Persia, Ultima, and so many more.

Ah well, yeah, while i played a few older games i pretty much missed the great first wave of gaming. But i was RIGHT THERE in the GOLDEN AGE. Early 90'ies... awesome stuff.

Excluding Educational softwares on my primary school's RM Nimbus 186 [Aaaaah, Through the Dragon's Eye!] The first game I remember playing for any length of time was Jetpack on the ZX Spectrum. Damn, someone said they felt old for playing Flashback! It was years before I got an Amiga.

EDIT - Oh snap! Didn't read the OP properly!

Earliest PC gaming memories didn't come 'til much later on, I suppose I'd have to go with Star Trek - A Final Unity.

Moto Racer was amazing, it was also the first game I've ever played.

Never again have i seen such a fun motorcycle racing game, although i throughly enjoyed the motorcycles in the midnight club series.

Exile: Escape from the Pit
Flashback: Quest for Identity.

Fuck. I feel old.
Me too:

The Secret of Monkey Island
Quarantine
Maniac Mansion
Alone in the Dark
Stunts
Blackthorne
Out of This World
Shadow of the Comet

Those were only the first batch of "good" PC games (not counting the merely "ok" ones) I played on my state-of-the-art 386, after years of being a console gamer. And don't even get me started on the MSX, or I'll REALLY get nostalgic on you.
