To me that would be Curse of Monkey Island. Name me another game that had a pirate musical number, some 4th walls breaking (ok there other games that done it), using a banjo as a guitar and the insult battle!

Also runner up does to No More Heroes.

I like all the Monkey Island Games, but the funniest game I've played is Leisure Suit Larry: Magna Cum Laude.

The absolute funniest is a (white) trash game called Country Justice: Revenge Of The Rednecks (?) which actually tries to be funny, but then it's so hilariously bad you'll laugh your head off in the first 5 minutes. That game is un-fucking-believable, I seriously don't understand how Big Rigs got so popular and this one not.

Anyway, Dungeons of Dredmor is pretty histerical too. One day my game crashed on start, and then an achievement popped up saying "Suddenly, the dungeon collapses!" Man I fell down from my chair laughing at that; probably the happiest game crash of my life. Then, there are some funny ingame elements such as "Congratulations! You died!" which never gets old, and an enemy character simply named "Unfriendly AI" which looks like a Protectron from the Fallout games... sorta.

Other funny games include Wolfschanze (same reasons as Country Justice), Sniper (or Snajper, some european game), Eat Lead: The Return Of Matt Hazard ("I'll soak your ass!") and Super Meat Boy for its cutscenes.

Armed and Dangerous. This thread just jogged my brain and I remember constantly playing this on my friends Xbox and getting a ridiculous amount of laughs out of it.

Fable 3 was funny (even if the rest of it was terrible), and all Bioware games make me chuckle.
I haven't played portal 2 yet, but when I do I'm sure it'll become the new favourite.

For me the most intentionally funny game I've ever played was Comic Jumper. The most unintentionally funny one was probably Final Fantasy 13.

Portal, Dungeons of Dredmor (masterfully done game, i might add), GTA4 (had a lot of fun with friends playing it), and Saints Row 2. All games that are awesome and hilarious in their own way. Gotta love 'em.

I'm gonna have to for with Portal 2 as well. I've heard some funnier jokes in other games but Portal 2 is consistently hilarious.

Agreed, the entire series was spectacular and I can never thank Insomnaic games enough for anything they do, even if Resistance tended to disagree with me a little bit.

More recently, Katawa Shoujo has proven itself to be an eccentric yet brilliant comic masterpiece. (As if anything conjured up by a developer made entirely out of 4channers WASN'T going to be completely full of awkward sexual moments and 'in-jokes'). The game, although it's more of a book, can also be labelled as 'emotional and deep', at times rolling a single tear down my quivering face. I am surprisingly attached to it, despite having a usual adversity to anything that can be described as either 'anime' or 'manga'.

It's a tie between Portal 2 and the Ace Attorney series. Portal 2 is more quotable, but the Phoenix Wright games sometimes crack me up with character design alone.

Portal 2, Monkey Island and Orcs Must Die! The last one wasn´t was funny as the first one, but funny nonetheless. Gotta love that apprentice.
Nothing else comes to mind at the moment.

It's gotta be Mystical Ninja on the N64. What a hilarious game. I'm not quite sure if it was supposed to be, but a combination of ridiculous characters and plot lines mixed with some very 'slap-dash' translation usually always gets a few lols.
