In recent history easily the Portal series. I also thought Brutal Legend was pretty funny.

Overall probably either Monkey Island or the Space Quest/Quest for Glory series-es. Space Quest has hilarious deaths at every turn.

Fallout 2 without a doubt. I couldn't stop laughing the time you make a guy chop off his little finger, because you said it was how you signed a contract and you reply "You could dip it in garlic sauce".

Saints Row 3. I laugh every time I say the word penetrator as I see my badass character holding it and beating people to death with it.

DeathSpank was legitimately hilarious in so many ways. It's rare for games to have sight gags in addition to hilarious dialogue, but it had plenty. It even tossed in a few parodies & references. But what else would you expect from Ron Gilbert?

Yeah, I'd go for most ratchet and clank games on the first time around.

Portal and Portal 2, however, I feel has more sustainability. That is, it stays funnier on the 2nd 3rd etc. playthrough.

For sure.The sheep line at the end of the second one still gets me.

I do not understand why people think that game is funny. Is it nostalgia, from when they played it as a kid and thought that swimming in shit was hilarious? I loved that game. I think it was the first one I played that effectively broke the 4th wall. I remember in the tutorial, an old guy was saying "Jump by pressing the Triangle Button."
To which the protagonist replied, "Button? What button? But alright. I'll humour you, old man."
I thought it was funny.

Also, Assassin's Creed: Revelations had some parts I thought were pretty funny. There's a part where you have to knock out a few minstrels and take their clothes. The absurdity of the situation struck me as I was doing it, and I just started laughing. And later, as Ezio is pretending to be a minstrel, he's playing the lute horribly and singing retarded songs. And if I'm not mistaken, there's a recruitment mission where you have to hit up a fight club. The challenger/ potential recruit insults you to rile you up, to which Ezio calmly replies, "If I had a message for you, I'd pass it on through your mother."

Sam and Max, Monkey Island, Day of the Tentacle, Zak McKracken and the Alien Mindbenders, and Grim Fandango rate highly, but I'm just going to go ahead and throw MDK and MDK2 out there.

It was a shame that Saints Row The Third did not come close to the epicness of Saints Row 2.
I agree with this very much. The courtroom scene near the very beginning of the game with Johnny Gat was surprisingly hilarious and it kept it up consistently for almost the entire game. Not sure if it's my absolute #1 funniest game but it's very good either way.
