I'd lock a 1000 cats in my vault and leak radiation into the food and water and see what we had when the vault was opened. We'd either get Cat from Red dwarf, or scary ass mutated saber tooths.
Or just dead cats.
This is the Fallout universe we're talking about. Only half of everything dies, the other half mutates.

I would see the effects of first dividing the people into factions at the brink of war and then arming them and killing the faction leaders.

All the vault people play portal until they have completed the game. Then are put in a Vault with no weapons, but a real Glados, and a companion cube.

100 Vault Dwellers. 5 Deathclaws would be in the Vault. They would be domesticated pets for the Dwellers. 5 more Deathclaws, the killer kind, would be released after 10 years. The Dwellers would either slaughter the whole Deathclaw population out of fear or be slaughtered themselves.
Or they would convince the domesticated Deathclaws to kill the rabid ones

300 people in one vault. then after the first 5 years, every 6 months 100 people press a button on a board of 100. 33 of those buttons kill every one of their relatives.

It must have been suggested already, but...

Take a bunch of die-hard 360 owners who are addicted to Halo / CoD / Gears of War / Any "Real is Brown Ultraviolence" shooter you care to name.

The only source of entertainment in the Vault is Wii Sports.

vault 13: all types of metal are played constantly (but not to the extent that everyone goes deaf)
Vault 13 already exists - someone never played Fallout 1. :p

The experiment with Bunker 43 was that a Goat was made overseer, and the security staff had strict instructions to protect him. The Bunker was also home to a Hei Gui stealth suit, and one man who really hated goats...

We wrote a few of these a while back.

Put in two groups in a virtual war-zone (ie Americans v French). Give them laser-rifles and enough food to last until Vault opened. Let's see who wins!

Suppose this one has to be pitched...

SYNOPSIS: There is a large button in public display. Place considerable amounts of material stressing, very clearly, that NOTHING BAD WILL HAPPEN if the Button is pressed.

The Button is behind bullet-proof glass. Two years in, the glass automatically removes itself.

HYPOTHESIS: The presence of the Button, combined with the extreme propaganda of "nothing bad will happen", will most likely convince people something bad WILL happen. The button shall therefore be kept off-limits.

SYNOPSIS: As above, but the Button causes the entire Vault to self-destruct, killing all inhabitants.

HYPOTHESIS: I give it a week.

For thoughs who don't know, in Fallout several vaults where invented to keep people safe after the bombs fell. However, many of them were instead used for weird twisted experiments with the inmates. For instance, there was a vault in Fallout 3 full of nothing but Clones of this guy called Gary.

If you could design a vault experiment what would you do?

I'd lock a 1000 cats in my vault and leak radiation into the food and water and see what we had when the vault was opened. We'd either get Cat from Red dwarf, or scary ass mutated saber tooths.
Where's the vault with the clones? I haven't come across it...

I'd tamper with the holovid and intercom system so that whenever someone decides to watch a film or listen to the radio, hell, take any action whatsoever (i.e. opening a door) there's a chance that they'll be Rickrolled via a projector or the radio in their PipBoy.

I expect either an incredibly insular population or a society of paranoid schizophrenics to come about rapidly.
