It depends on the kind of Joker that you like. If you want a serious Joker, then Heath Ledger would be the best, but if you want a lighter, yet creepy Joker, then Mark Hamill is the best.

Are you kidding me? The Joker has been the most famous comic book villain for decades. The Joker was responsible for reviving the Superhero film genre when he appeared in Tim Burton's Batman.
Even if you haven't read the comics, you know who the Joker is.
