The Portal series for definite, the first game especially.

Timesplitters: Future Perfect, that game is very funny.

Battlefield Bad Company has some pretty good moments.

And pretty much any game featuring The Simpsons is funny.

Saints Row 2 has to be the funniest game I have ever played, +30 hours of random missions and side missions, totally badshit loco character creation and the cast of characters in the story.

It was a shame that Saints Row The Third did not come close to the epicness of Saints Row 2.

Oh and all the random mods for Warcraft III are also fun.

Portal 2 is amazing of course but I have to go with "Time Gentlemen Please!" for sheer density. Day of the Tentacle was also amazing.

I've got to say if you play Moonbase Alpha with a couple of friends that it never disappoints, Microsoft Sam + Moon is a brilliant idea. I've had some great moments in GMod including a contraption of a pod, boosters and those gravity orbs, madness! I actually found Duty Calls (the Bulletstorm thing) fairly funny just because it was so blunt and right about everything CoD, textures were a mess, very facist, scripted and short etc. It was overdoing it a bit but it made it all the more funny for me.

All the games mentioned above are great (Monkey Island, SLH, Portal). However, the game that made me laugh the most when I was playing it has got to be Strong Bad's Cool Game for Attractice People. I mean, come on - Uzi-Bazooka, my username, is a character from the fourth episode of the game. Of course, I've always been a huge fan of Homestar Runner, so its random, bizarre humor may not be for everyone.

I don't know about funniest, but Day of the Tentacle is defiantly up there.
Laverne (after thawing out a frozen hamster using a microwave and then facing the camera/player): "That was fun, but only because this is the 22nd Century. People who put hamsters in microwave ovens in my century get taken away from their parents and put up for adoption, so DON'T DO IT!"

Portal and Portal 2...right done that and now moving on.

The old point and click games had humour as a focal point of their writing and as such were often very funny, Monkey Island comes to mind.

More recently I've found myself actually laughing out loud to the dark side smuggler dialogue in The Old Republic. Ok maybe not the funniest thing I've ever heard in a computer game but still surprisingly funny.
