The best one is Bruno [http://www.baldwinpage.com/jan196.html]. It's good to start with, but once it gets goin, after about one hundred strips or so, it gets amazing. There's not a shred of doubt in my mind that this is the best webcomic ever.
My favourite webcomic would have to be Brawl In The Family, a Nintendo based webcomic centered mainly around the Kirby series, that'd be my pick for humour, as for other...
...yeah, I don't have any of those, all the webcomics I read are humour
The Adventures of Dr McNinja has always been a perennial favourite, although the latest mega-time-travel arc is wearing on a bit too much for me. The older stuff is still gold.
The Order of the Stick is great when it updates, which is not as frequently as i'd like, but it still has a great blend of comedy and story.
Honourable mentions to XKCD, Axe Cop, QC and The Gutters.

Dr McNinja. Best comic evar.
Honourable mention to Schlock Mercenary, Sluggy Freelance, XKCD, Looking For Group, Order of the Stick, Penny Arcade and Darths & Droids. I read QC for a long time, then lost my bookmark and cba to find the comic again. Being out of synch with a comic like that just makes it seem... crap. Maybe it always was...
Either Questionable Content or Lackadaisy Cats, both are great.
I used to read Least I Could Do and Looking For Group but got tired of them. LICD because the main character is just getting worse and LFG's story just doesn't seem to make sense anymore.
For me Goblins is king, followed closely by XKCD. QC is also funny at times so I keep reading it, but can't honestly say it's great. Order of the Stick is very good, but doesn't update often enough to check regularly.
xkcd is my current number 1, but the best webcommics on the web are the older, one page, Dresden Codaks. Not even xkcd is that absurd and that brilliant.
